abstract

veritable identification and differentiation of avian species is a vital step in conservative, taxonomic, forensic, legal and other ornithological interventions. therefore, this study involved the application of molecular approach to identify some avian species i.e. chicken (gallus gallus), muskovy duck (cairina moschata), japanese quail (coturnix japonica), laughing dove (streptopelia senegalensis), and rock pigeon (columba livia). genomic dna was extracted from blood samples and partial sequence of themitochondrial cytochrome b gene (358 bp) was amplified and sequenced using universal primers. sequences alignment and phylogenetic analyses were performed by clc main workbench program. the obtained five sequences were deposited in genbank and compared with those previously registered in genbank. the similarity percentage was 88.60% between gallus gallus and coturnix japonica and 80.46% between gallus gallus and columba livia. the percentage of identity between the studied species and genbank species ranged from 77.20% (columba oenas and anas platyrhynchos) to 100% (gallus gallus and gallus sonneratii, coturnix coturnix and coturnix japonica, meleagris gallopavo and columba livia). amplification of the partial sequence of mitochondrial cytochrome b gene proved to be practical for identification of an avian species unambiguously.

Genetic Diversity and Molecular Phylogeny of Iranian Sheep Based on Cytochrome b Gene Sequences

Phylogenetic relationships and genetic variation between two Iranian sheep breeds were analyzed using cytochrome b (cyt-b) gene sequences. The genomic DNA was isolated by salting out method and amplified cytochrome b gene using polymerase chain reaction restriction (PCR) method with a pair of primer. A partial sequence of cyt-b gene of Iranian sheep is 780 bp and contained 13 variable sites and...

Molecular Characterization and Phylogeny Analysis Based on Sequences of Cytochrome Oxidase gene From Hemiscorpius lepturus of Iran

Abstract: Background: Hemiscorpius lepturus is a medically important scorpion found along the Iranian borders, especially near to Khuzestan Province in the south-west of Iran. This is the only non-buthid scorpion which is potentially lethal in southern Iran and is responsible for severe dermonecrotic scorpionism. OBJECTIVES: In this study, DNA fragment of the mitochondrial cytochrome c oxidase ...

Molecular phylogeny of the chipmunks inferred from Mitochondrial cytochrome b and cytochrome oxidase II gene sequences.

There are currently 25 recognized species of the chipmunk genus Tamias. In this study we sequenced the complete mitochondrial cytochrome b (cyt b) gene of 23 Tamias species. We analyzed the cyt b sequence and then analyzed a combined data set of cyt b along with a previous data set of cytochrome oxidase subunit II (COII) sequence. Maximum-likelihood was used to further test the fit of models of...

Sequence Analysis of the Cytochrome b Gene Trimorphomyces papilionaceus Mitochondria

The DNA sequence of the cytochrome b (rob) gene region of Trimorphomyces papilionaceus mitochondrial DNA has been determined. The cob gene is interrupted by an intron of 1267 bp, which has an open reading frame of 897 bp contiguous to the upstream exon. The intron belongs to group IB and contains a reading frame with a GIY-10-YVG motif. The deduced amino acid sequence shows 52.-40% homology wit...

Geographic Distribution of Leishmania Species in Ecuador Based on the Cytochrome B Gene Sequence Analysis

A countrywide epidemiological study was performed to elucidate the current geographic distribution of causative species of cutaneous leishmaniasis (CL) in Ecuador by using FTA card-spotted samples and smear slides as DNA sources. Putative Leishmania in 165 samples collected from patients with CL in 16 provinces of Ecuador were examined at the species level based on the cytochrome b gene sequenc...
